P21 (P021) — synthetic neurogenic peptide based on the biologically active region of human ciliary neurotrophic factor (CNTF, residues ~148–151), to which an adamantane amino acid was added to increase stability and lipophilicity (Li et al., FEBS Lett 2010, PMID 20600002). P21 is conceptually related to Cerebrolysin's neurotrophic approach but is not a fragment of it. The adamantane base was added to increase lipophilicity and blood-brain barrier permeability (preclinical data indicate sufficient brain penetration; no human pharmacokinetics) and to protect the molecule from enzymatic degradation — making P21 suitable for peripheral administration (preclinical work mostly used the oral route).
