Nootropics Booster is a synthetic peptide, ac-Semax-NH2 (an acetylated, amidated form of Semax). Semax is a Russia-developed neuropeptide derived from the ACTH fragment 4-7, modified with acetyl and amide groups to improve resistance to enzymatic degradation and lipophilicity (no direct pharmacokinetic data for this form). Unmodified intranasal Semax was studied in Russian (mostly non-randomized) protocols for stroke and traumatic brain injury; in Ukraine the Semax marketing authorizations lapsed (2009/2014), and Russian-made medicines are subject to MOH restrictions.

In preclinical studies (mostly in animals, unmodified Semax), potential was described for supporting memory, attention and information-processing speed, neuroprotection from oxidative stress, cortisol regulation and BDNF increase. These effects are not proven for the acetylated ac-Semax-NH2 form and have not been established in humans in controlled trials.

Nootropics Booster (ac-Semax-NH2) — an acetylated, amidated form of the Semax peptide derived from the ACTH(4-7) fragment (Pro-Gly-Pro). Hypothesized mechanism (from preclinical studies of unmodified Semax): stimulation of BDNF synthesis — a neurotrophin regulating axon growth, synaptic plasticity and neuron survival (Dolotov et al., J Neurochem 2006 — in rats, BDNF rose in the basal forebrain 3 h after a single intranasal dose). Increased NGF/trkB activity and reduction of excess nitric oxide under ischemia have also been described (Bashkatova et al., Brain Res 2001) — a neuroprotective effect. The acetyl and amide modifications are intended to improve resistance to enzymatic degradation and lipophilicity; there is no direct pharmacokinetic data (including blood-brain-barrier penetration) for this form. Important: these data are for unmodified Semax (mostly animal work and non-randomized Russian stroke studies, e.g. Gusev 1997, n=30 vs 80 controls); for the acetylated ac-Semax-NH2 form itself there are no in vivo preclinical or clinical data (Nicula et al., HVM Bioflux 2026), and clinical efficacy in humans is not established.
